Core Responsibilities

Primary Functions

  • Plan, set up, and execute mammalian cell culture workflows (adherent and suspension) for transient and stable recombinant antibody expression, maintaining aseptic technique, cell line health, and passage records to support reproducible antibody production.
  • Run hybridoma generation and maintenance processes where applicable, including cell fusion, cloning, screening, expansion and cryopreservation, while documenting clone lineage and performance metrics.
  • Perform small- and large-scale upstream expression campaigns in shake flasks, spinner bottles and benchtop bioreactors; monitor culture conditions (viability, density, metabolites) and make adjustments to optimize yield and quality.
  • Execute downstream purification processes including protein A/G affinity chromatography, ion exchange, size exclusion and buffer exchange (tangential flow filtration/dialysis) to produce research- and clinical-grade antibodies with documented purity and recovery statistics.
  • Characterize antibody products with analytical assays such as SDS-PAGE, Coomassie or silver staining, Western blot, UV spectrophotometry, and concentration/purity calculations; generate interpretive lab reports and trending summaries.
  • Develop and run immunoassays (indirect/capture ELISA, competitive ELISA), optimize coating and blocking conditions, perform titrations and endpoint analyses for antibody binding and titer determination.
  • Perform functional and binding characterization using flow cytometry, surface plasmon resonance (SPR), biolayer interferometry (BLI), and affinity/kinetics assays to assess specificity, cross-reactivity and affinity of antibodies.
  • Execute antibody labeling and conjugation (fluorophore, HRP, biotin, enzyme) using validated chemistries; purify labeled antibodies and characterize degree of labeling, activity retention and stability.
  • Conduct stability studies and accelerated degradation experiments on antibody preparations; prepare samples, monitor change in activity or aggregation, and document results per study protocol.
  • Prepare, aliquot, and maintain reagent inventories including buffers, media, sera, growth supplements and chromatography resins; prepare stock solutions and quality-check incoming reagents per SOPs.
  • Maintain accurate, contemporaneous laboratory documentation including electronic or paper lab notebooks, batch records, SOPs and QC checklists to satisfy GLP, GMP or internal quality requirements.
  • Troubleshoot routine process and assay issues (low yield, contamination, inconsistent assay signal), perform root cause analysis, implement corrective actions and escalate complex problems to senior staff or principal investigators.
  • Participate in method transfer and scale-up activities, supporting transition of bench protocols to pilot or production scale while documenting process parameters, critical quality attributes and process capability.
  • Support generation of lot release documentation and assist QC teams with testing required for release criteria, stability, and traceability of antibody batches.
  • Maintain and calibrate laboratory equipment used for antibody development (centrifuges, biosafety cabinets, incubators, chromatography systems, plate readers, flow cytometers) and coordinate preventative maintenance and service requests.
  • Assist with training and mentoring of junior technicians and interns on cell culture, aseptic technique, assay execution, and laboratory safety procedures, ensuring consistent competency and compliance.
  • Implement and follow laboratory safety, biosafety and waste disposal procedures, including handling of biohazardous materials, chemical safety, and spill response protocols to maintain a compliant lab environment.
  • Collaborate cross-functionally with research scientists, process development, analytical, and quality teams to support experimental design, data interpretation and continuous process improvement initiatives.
  • Prepare and present experimental results, assay validations and technical summaries to project teams and stakeholders, supporting decision-making for lead selection and downstream development.
  • Support sample management and chain-of-custody for biological samples and antibody stocks; process incoming/outgoing samples and maintain accurate LIMS or inventory entries.
  • Participate in project planning and timelines, estimate resource needs for antibody campaigns, proactively manage competing priorities and communicate project status updates to supervisors.
  • Conduct hygiene and sterility monitoring, environmental monitoring and implement contamination control measures to protect cultures and maintain data integrity across antibody workflows.
  • Stay current with antibody engineering and analytical technologies (next-gen sequencing for B-cell repertoires, affinity maturation platforms, novel labeling chemistries) and recommend adoption where it accelerates program goals.

Secondary Functions

  • Prepare routine lab reagents, buffers and media and maintain batch preparation logs and QC checks to ensure consistency across experiments.
  • Coordinate ordering and vendor interactions for specialized antibodies, chromatography resins, disposables and cell culture reagents; maintain supplier qualification records where applicable.
  • Assist in drafting, reviewing and updating SOPs, work instructions and protocol templates to reflect process improvements and regulatory expectations.

Education & Experience

Educational Background

Minimum Education:

  • Bachelor’s degree (B.S.) in Biology, Biochemistry, Molecular Biology, Immunology, Bioengineering, or a closely related life science field.

Preferred Education:

  • Bachelor’s plus 2–4 years of relevant antibody development or bioprocessing experience, OR
  • Master’s degree (M.S.) in Immunology, Biochemistry, or Biotechnology with hands-on lab experience.
